Data Sources & Evidence Base
Oocyte yield Fanton et al. 2023 (Fertil Steril) — SART CORS 2014–2019, n=402,411 retrieval cycles. Strong positive linear correlation between AMH, age, and oocytes retrieved. AMH and age together predict oocyte yield; age alone dominates euploidy.
Blastocyst arrest Russo et al. 2025 (Aging) — 25,974 embryos from 1,928 cohorts. Developmental arrest rate increases with age: 33% (<35), 38% (35–37), 40% (38–40), 44% (41–42), 44%+ (>42). Arrest correlated with meiotic aneuploidy.
Euploidy rate PMC 2022 (Reprod Sci) & ART Calculator (Polyzos et al. 2019, Front Endocrinol) — Age is the sole significant predictor of euploidy rate (p<0.0001). AMH and oocyte number do not independently affect euploidy per biopsied blastocyst. ART Calculator validated in 1,547 ICSI/PGT-A cycles (r=0.91).
Euploid LBR SART CORS 2014 (Fertil Steril 2021) — Live birth rate from euploid blastocyst FET is ~60–65% in published SART data and does not vary significantly with infertility diagnosis or age once embryo is confirmed euploid. A conservative 50% figure is used here to account for real-world variation, patient age effects on implantation, and uterine factors not captured in population data. Estimates reflect a single euploid blastocyst SET.
Fertilisation SART CORS national data & clinical consensus — MII rate ~78–80%; ICSI fertilisation (2PN) rate ~75–80% of MII; conventional IVF ~65–75% of MII. Surgical sperm associated with lower fertilisation and blastocyst rates (ART Calculator, Polyzos 2019).
PGT-M genetics Mendelian inheritance — AR: 25% affected, 75% unaffected (carrier + non-carrier). AD: 50% affected, 50% unaffected. XLR: all female embryos unaffected; 50% of male embryos affected — effectively ~75% suitable for transfer. Rates are theoretical Mendelian probabilities applied to euploid blastocyst cohort.
PGT-SR rates Zhou et al. 2024 (BMC Pregnancy & Childbirth) — 4-year real-world retrospective cohort, 1,034 PGT-SR cycles, 4,129 biopsied blastocysts. Euploidy rates: reciprocal translocation 30.1%, Robertsonian translocation 46.1%, inversion 57.3%. These real-world figures are more conservative than theoretical meiotic segregation models, reflecting additional de novo aneuploidies on non-translocation chromosomes. Rates vary significantly by specific translocation and breakpoints.
